Vectors that integrate the transferred DNA into the chromosomes of the recipient cell have the greatest potential for long term expression. Retroviral vectors and adeno-associated viral yectors have integrating functions. The persistence of the transgene DNA in the genome of the recipients cell does not, however, guarantee long term expression in that cell. The production of the intended mRNA and protein may decline because of inactivation of the transgene promoter even though the DNA persists. (Bestor, 2000)
